A custom field needs to show a dropdown of all users assigned to a specific permission set/role, so a task can be assigned to a specific person — using a static Enumeration would work but would require manually maintaining the list as roles/staff change. Is there a way to have this list populate dynamically instead?

Answer

Use a Reference-type custom field instead of an Enumeration — a Reference points to a real, always-current data source (like the standard user/employee data), so it doesn't need manual list maintenance the way a static Enumeration would. In the Reference field's List of Values dialog, apply a Saved Search to narrow the results down to just the users who actually hold the relevant permission set/role, and set that Saved Search as the default in the Base Profile so it automatically applies whenever the LOV opens for the affected users — this gives you an effectively self-maintaining, role-scoped dropdown without needing to manually update an Enumeration's fixed value list over time.
