Is it possible to use the CTRL+K lookup shortcut on a custom field, for example to open a value list for a custom field linked to an enumeration?

No — as currently implemented, CTRL+K lookup isn't supported for custom fields in general, including ones linked to an enumeration. This is a limitation of the custom field framework itself, not a missing configuration step on a specific field.
