When creating a new Inventory Part, is it possible to have the Part Number field auto-populate (e.g. from an Oracle sequence) instead of requiring it to be typed in manually every time?

Yes, if parts are created starting from Engineering rather than directly as an Inventory Part: go to Engineering Parts, create a new record with only the Part Description filled in, leave the Part Number field empty, and save — this auto-generates the next available sequential part number for the new Engineering Part. From there, use the Engineering Transfer (or Create Inventory Part) functionality to carry that engineering part, with its auto-generated number, over into an actual Inventory Part.
