As of the versions this was last checked against, no — editing the URL on an already-created ExternalNavigationLink command button isn't supported; it wasn't prioritized for near-term releases either. If you need to change the target URL, recreating the command button with the new URL is the workaround. Since this is the kind of gap that does eventually get addressed, check current release notes in case it's since been implemented.
